The Thermo Scientific™ Endonuclease IV (Endo IV) is a recombinant DNA repair enzyme derived from E. coli, specialized in recognizing and processing apurinic/apyrimidinic (AP) sites in double-stranded DNA. By cleaving the phosphodiester bond 5′ to AP lesions, it generates a 3′-hydroxyl group suitable for DNA polymerase extension, making it a key tool for DNA repair studies.

In addition to AP endonuclease activity, Endo IV functions as a 3′-diesterase, removing damaged 3′-blocking groups such as 3′-phosphoglycolate and 3′-phosphate. It also exhibits 3′→5′ exonuclease activity, preferentially processing substrates with 3′-recessed ends. The enzyme operates independently of Mg²⁺ and remains active in the presence of moderate EDTA concentrations, ensuring flexibility in diverse experimental conditions.

    Key Features

    • AP Site Recognition & Cleavage – Specifically cleaves at apurinic/apyrimidinic sites in dsDNA.
    • Multi-Activity Enzyme – Functions as an AP endonuclease, 3′-diesterase, and 3′→5′ exonuclease.
    • Damage Processing – Removes 3′-phosphoglycolate and 3′-phosphate groups from damaged DNA ends.
    • Metal-Independent – No Mg²⁺ required; remains active even in the presence of EDTA.
    • Research-Grade Purity – Suitable for high-precision DNA damage and repair studies.

    Applications

    • DNA damage and repair research
    • Single-cell electrophoresis (comet assay)
    • Antitumor drug mechanism studies
    • DNA structure and repair pathway analysis
    • SNP analysis and mutation detection
