By cleaving the phosphodiester bond 5′ to AP lesions, it generates a 3′-hydroxyl group suitable for DNA polymerase extension, making it a key tool for DNA repair studies.
In addition to AP endonuclease activity, Endo IV functions as a 3′-diesterase, removing damaged 3′-blocking groups such as 3′-phosphoglycolate and 3′-phosphate.
