Quantitative colorimetric assay for measuring total and oxidized glutathione in diverse biological samples
The Invitrogen™ Glutathione Colorimetric Detection Kit is a ready-to-use, research-use-only assay for quantitative measurement of total glutathione (GSH + GSSG), free GSH, and oxidized glutathione (GSSG) in whole blood, plasma, serum, erythrocytes, urine, cell lysates, and tissue samples. This species-independent assay features a simple, no-wash workflow and delivers sensitive, reproducible results using a microplate reader at 405–412 nm.
Glutathione is the most abundant non-protein thiol in mammalian cells, playing vital roles in redox homeostasis, detoxification, protein and DNA synthesis, and cellular defense against oxidative stress. Accurate quantification of GSH and GSSG is essential for oxidative stress, metabolism, and toxicology research.

Assay Principle
- Total GSH Measurement – The free thiol group of GSH reacts with the kit’s colorimetric substrate to form a highly colored product measured at 405 nm.
- GSSG Measurement – 2-vinylpyridine treatment blocks free GSH, allowing selective measurement of oxidized GSSG.
- Free GSH Calculation – Obtained by subtracting GSSG (converted to GSH equivalents) from total GSH.
Results can be obtained via endpoint detection or by kinetic measurement of color development.
