The Syphilis ELISA is an indirect immunoenzyme assay designed for the qualitative detection of antibodies against Treponema pallidum in human serum or plasma. This test is intended for the laboratory diagnosis of syphilis and can be used in both routine and automated ELISA systems.
The assay employs Treponema pallidum antigens adsorbed onto the surface of polystyrene wells, enabling the detection of specific IgG and/or IgM antibodies. Results are visualized through a colorimetric reaction using TMB substrate, ensuring high sensitivity, specificity, and reproducibility.

Principle of the Test
- Antibodies present in the patient serum or plasma bind to Treponema pallidum antigens coated on the polystyrene surface.
- Unbound immunoglobulins are washed away.
- An enzyme-labelled anti-human globulin is added, binding to the antigen-antibody complex.
- After washing, TMB substrate develops a blue colour, which turns yellow upon addition of the acid stop solution.
- The resulting optical density is proportional to the concentration of antibodies against Treponema pallidum.
