It serves as a redox indicator in selective and differential culture media to visually distinguish microbial growth. The reagent is colorless in its oxidized form and converts into red insoluble triphenylformazan upon reduction by many bacteria and fungi, providing a clear and persistent color change.

Preparation & Use:
- Preparation:
- Refer to the technical sheet of the base medium being used (e.g., Slanetz and Bartley Agar Base, Azide Maltose Agar KF, etc.).
- Use one tube of TTC 1% Supplement (10 mL) per 1 liter of the medium.
- Procedure:
- Mix thoroughly after adding the supplement to the base medium.
- Pour the prepared medium into Petri dishes or tubes as required.
Quality Control:
- Visual Inspection:
- The solution should be clear and colorless.
- Microbiological Testing:
- Prepare plates with Slanetz and Bartley Agar Base supplemented with TTC 1%.
- Incubate plates at 36 ± 1°C for 48 hours.
- Results:
- Enterococcus faecalis (ATCC 29212): Good growth; red colonies.
- Escherichia coli (ATCC 25922): Growth inhibited.
