The Human 8-Hydroxy-deoxyguanosine (8-OHdG) ELISA Kit is a sensitive and quantitative enzyme-linked immunosorbent assay designed for the measurement of 8-OHdG, a widely recognized biomarker of oxidative DNA damage. 8-OHdG is formed when reactive oxygen species (ROS) attack guanine bases in DNA, making it a critical indicator in studies of oxidative stress, aging, cancer, inflammation, neurodegeneration, cardiovascular diseases, and environmental toxicity.
This kit enables accurate quantification of human 8-OHdG levels in serum, plasma, urine, culture media, cell lysates, tissue homogenates, and other biological fluids, supporting a broad range of life-science and biomedical research applications.
Intended Use
This ELISA kit is intended for the quantitative detection of Human 8-OHdG antigen in biological samples for research purposes only.
Assay Principle
The assay is based on a solid-phase sandwich ELISA technique. Microplate wells are pre-coated with a high-affinity antibody specific for Human 8-OHdG. During the first incubation, 8-OHdG present in standards or samples binds to the immobilized antibody.
After washing to remove unbound materials, an HRP-conjugated secondary antibody specific to Human 8-OHdG is added, forming a stable antibody–antigen–enzyme complex. Excess conjugate is removed through additional wash steps.
The enzymatic reaction is initiated by the addition of TMB substrate, producing a blue color that is proportional to the amount of bound 8-OHdG. The reaction is stopped with an acidic stop solution, changing the color to yellow. Absorbance is measured at 450 nm using a microplate reader, and sample concentrations are calculated by comparison with a standard curve.

Storage & Stability
