Originally developed at the Roswell Park Memorial Institute for culturing human leukemic cells, RPMI 1640 has become one of the most versatile and extensively utilized media in immunology, oncology, and cell biology research.

This powder formulation contains L-glutamine and phenol red, supporting optimal cell metabolism and enabling visual monitoring of pH changes during culture. It is supplied without HEPES and sodium bicarbonate, allowing researchers to customize buffer systems and prepare the medium according to specific experimental requirements. The powder format offers flexibility, extended shelf life, and cost-effective preparation of large volumes of culture medium.

RPMI 1640 is distinguished by its unique composition, including high concentrations of vitamins such as inositol and choline, and the presence of essential components such as biotin, vitamin B12, and para-aminobenzoic acid (PABA), which are not found in many other basal media. It also contains the reducing agent glutathione, supporting cellular health and enhancing viability in sensitive cell cultures. Because the medium does not contain proteins, lipids, or growth factors, supplementation with serum, typically 10% fetal bovine serum (FBS), is required to support optimal cell growth.

This medium is suitable for culturing a wide variety of mammalian cell types, including HeLa, Jurkat, MCF-7, PC-12, peripheral blood mononuclear cells (PBMC), astrocytes, and carcinoma cells.
