The Invitrogen™ Goat anti-Human IgM Secondary Antibody, HRP-conjugated (Catalog #31415) is a polyclonal, affinity-purified antibody specifically designed for the detection of human IgM (Fc5μ fragment) in immunoassays. This antibody is conjugated to Horseradish Peroxidase (HRP) to enable enzyme-based signal detection in applications such as Western blot (WB), Immunohistochemistry (IHC), and Immunocytochemistry/Immunofluorescence (ICC/IF).
With exceptional sensitivity, minimal cross-reactivity, and broad dynamic range, this secondary antibody is ideal for detecting human IgM heavy chains in complex protein mixtures using colorimetric or chemiluminescent detection systems.
Key Features & Benefits
✅ Specific for Human IgM Heavy Chain (Fc5μ Region)
- Selectively binds the Fc5μ portion of human IgM.
- Demonstrated no cross-reactivity with human IgG or IgA in electrophoresis-based testing.
- May cross-react with IgM from other species—validate if needed.
✅ Conjugated to HRP for Enzyme-Based Detection
- Enables colorimetric, chemiluminescent, or fluorescent detection using compatible substrates (e.g., TMB, DAB, ECL).
✅ Affinity-Purified for High Specificity
- Purified using affinity chromatography for low background and high signal clarity.
✅ Lyophilized for Long-Term Stability
- Supplied lyophilized; reconstitute with 2.0 mL distilled water to yield 0.8 mg/mL working concentration.
- Extended shelf life when stored properly.

Restoration & Storage Instructions
- Reconstitution: Add 2.0 mL of distilled water to lyophilized vial to obtain a final concentration of 0.8 mg/mL.
- Allow the solution to stand 1–2 hours at room temperature, then centrifuge if solution is not clear.
- Short-term storage: Store undiluted at 4°C for several weeks.
- Long-term storage: Add an equal volume of glycerol (final 50%) and store at –20°C.
- Post-dilution usage: Use immediately—do not store diluted antibody for extended periods.
Target Information – Human IgM
- Immunoglobulin M (IgM) is the first antibody isotype produced during an immune response and plays a vital role in early immunity.
- It exists primarily in a pentameric form in the bloodstream and is critical in neutralizing pathogens, activating complement, and clearing antigens.
- The Fc5μ portion is a unique region of the IgM heavy chain, enabling selective detection of IgM without cross-reactivity to IgG or IgA.
Compatible Detection Systems
-
HRP Substrates:
- TMB (3,3′,5,5′-Tetramethylbenzidine) for ELISA
- DAB (3,3′-Diaminobenzidine) for IHC
- ECL (Enhanced Chemiluminescence) for Western blot
- Compatible with a variety of membranes (PVDF, nitrocellulose) and imaging systems.
Why Choose This Antibody?
✅ Highly specific to human IgM (Fc5μ)—no cross-reactivity with IgG or IgA
✅ Versatile across multiple applications: WB, IHC, ICC/IF
✅ HRP conjugation supports sensitive enzymatic detection methods
✅ Reliable performance in immunoassays with low background noise
✅ Backed by Invitrogen quality and validation data
This antibody is ideal for researchers studying primary immune responses, B cell activity, autoimmune conditions, and human IgM protein expression using HRP-based detection.
