The Invitrogen™ CD184 (CXCR4) Monoclonal Antibody (12G5), APC-conjugated is a high-affinity, flow cytometry–validated antibody targeting human CXCR4 (CD184), a chemokine receptor involved in immune signaling, HIV entry, and developmental biology. The 12G5 clone binds to an extracellular epitope of CXCR4, enabling specific detection of surface-expressed CXCR4 on lymphocytes and other immune cell populations.
CXCR4 is a seven-transmembrane G-protein-coupled receptor for stromal-derived factor-1 (SDF-1/CXCL12), and acts as a co-receptor for X4-tropic HIV-1 and HIV-2. Its expression is rapidly upregulated upon immune stimulation and plays a critical role in hematopoiesis, neurogenesis, inflammation, and tumor metastasis.
Key Features and Benefits
- Flow cytometry validated: Optimized and pre-titrated for use in multicolor flow cytometry panels.
- Clone 12G5: Highly cited monoclonal antibody with published specificity for human CXCR4.
- APC-conjugated: Excitable with red laser; ideal for panels requiring far-red fluorophores.
- Inhibitory activity: Reported to block HIV-1 entry and SDF-1–induced signaling.
- Highly specific: Minimal cross-reactivity; suitable for profiling CXCR4 in human PBMCs.

Biological Background
CXCR4 (CD184) is a chemokine receptor essential for directing the migration of hematopoietic, immune, and neuronal cells. It binds to CXCL12/SDF-1, initiating signaling pathways that regulate cell movement, calcium mobilization, and MAPK activation. CXCR4 plays critical roles in cardiac development, vascular remodeling, and immune cell trafficking. It is a coreceptor for HIV-1 (X4-tropic strains) and a therapeutic target in cancer and inflammation.
The 12G5 antibody has been shown to:
- Partially inhibit SDF-1–induced chemotaxis and calcium signaling
- Block HIV-1 and HIV-2 envelope-mediated fusion
- Stain CXCR4 on T cells, especially naïve subsets in human peripheral blood
The Invitrogen™ CD184 (CXCR4) Monoclonal Antibody (12G5), APC-conjugated, is a powerful tool for detecting, quantifying, and characterizing CXCR4 expression in immune, developmental, and HIV research. Its red-fluorescent conjugation and proven performance in flow cytometry make it ideal for multicolor immune phenotyping, disease modeling, and therapeutic target validation.
