Column Use & Maintenance
Initial Use & Conditioning
- Flush new columns with 15 column volumes of water before first use.
- Condition with a 40 mM EDTA.2Na solution (filtered, unadjusted pH) at low flow rate (20-24 hours).
- Alternatively, use a 0.2 M NaH₂PO₄ + 0.3 M sodium acetate solution for 2 hours.
Routine Use & Column Care
- Proteins elute with salt and/or pH gradients (pH 6-7 recommended).
- Filter mobile phases and samples to avoid clogging the column frit.
- Avoid prolonged exposure to pH above 8 to prevent degradation of the stationary phase.
- Store in 100% mobile phase A (short-term) or water (long-term at 4°C).
Column Regeneration & Longevity
- Invert column and flush with 40 mM EDTA.2Na overnight after every 250 injections.
- For heavy usage (500+ injections), replenish with bulk PolyCAT A™ material.
- Use at room temperature (20-25°C) to preserve column integrity.
