Invitrogen™ NuPAGE™ Antioxidant is a specialized electrophoresis reagent designed to maintain proteins in a reduced state during SDS-PAGE and protein transfer workflows. It is formulated to prevent sample reoxidation by preserving the reduced form of sensitive amino acid residues (such as methionine and tryptophan) throughout electrophoresis and blotting.

This reagent is used by adding it to the running buffer when working under reducing conditions with NuPAGE™ Bis-Tris or Tris-Acetate gels. It migrates with the proteins during electrophoresis, providing continuous protection from oxidation.

For optimal performance, use in conjunction with NuPAGE™ Sample Reducing Agent (NP0004) and other compatible reagents from the NuPAGE™ SDS-PAGE system. It is also recommended for addition to NuPAGE™ Transfer Buffer (NP00061) to preserve protein integrity during electroblotting.

Key Features and Benefits

  • Prevents protein reoxidation during electrophoresis and blotting
  • Maintains reducing environment throughout SDS-PAGE runs
  • Migrates with proteins to protect thiol-sensitive residues
  • Optimized for NuPAGE™ Bis-Tris and Tris-Acetate gels
  • Compatible with NuPAGE™ Sample Reducing Agent (DTT-based)
  • Improves resolution and transfer efficiency for sensitive proteins

Applications

  • SDS-PAGE under reducing conditions
  • Western blotting of reduced proteins
  • Protection of labile amino acids during electrophoresis
  • Enhancing transfer efficiency in blotting protocols

Usage Guidelines

  • Add NuPAGE™ Antioxidant to the running buffer when running reduced protein samples
  • Add to NuPAGE™ Transfer Buffer for enhanced protection during wet or semi-dry protein transfer
  • Use only with NuPAGE™ gel chemistries; not compatible with standard Tris-Glycine systems
