Intended Use
- To determine the MIC of Ampicillin-Sulbactam for Gram-positive and Gram-negative bacterial isolates.
- Particularly useful for testing beta-lactamase-producing bacteria, including Enterobacteriaceae, Pseudomonas spp., Staphylococcus spp., and Acinetobacter spp..
- Applicable in hospital microbiology laboratories, research facilities, and surveillance programs monitoring antimicrobial resistance.
Test Procedure
- Preparation:
- Prepare Mueller-Hinton Agar (or suitable medium for fastidious organisms) following CLSI or EUCAST guidelines.
- Allow the medium to solidify and ensure its surface is dry before inoculation.
- Inoculation:
- Prepare a bacterial suspension equivalent to 0.5 McFarland standard.
- Use a sterile swab to evenly streak the suspension across the agar plate.
- Application:
- Using sterile forceps, place an MTS™ Ampicillin-Sulbactam AMS strip onto the inoculated agar surface with the printed MIC scale facing upward.
- Incubation:
- Incubate the plate at 35 ± 2°C in an appropriate atmosphere (aerobic or with 5% CO₂ as needed) for 16–20 hours.
- Interpretation:
- After incubation, observe the inhibition ellipse. The MIC is the value where the bacterial growth intersects the strip.
- Interpret the results following CLSI or EUCAST breakpoints.
