Preparation Instructions

  1. Reconstitution: Suspend 56.0 g of dehydrated medium in 1 liter of distilled or deionized water.
  2. Dissolution: Heat to boiling while stirring gently until the powder is fully dissolved.
  3. Sterilization: Autoclave at 121°C for 15 minutes.
  4. Supplementation:
    • Cool to 45-50°C.
    • Aseptically add 2.5 ml of TTC 1% Supplement (ref. 80300).
    • Add 7 ml of Tergitol 1.5% Supplement (provided in the package).
    • Mix thoroughly.
  5. Dispensing: Pour into Petri dishes and allow to solidify.

  1. Sample Filtration:
    • Filter two 100 ml water samples through sterile membranes.
  2. Membrane Transfer:
    • Place each membrane onto the prepared Tergitol Agar Base plates.

    • Plate 1: Incubate at 37°C for 24 hours (for total coliforms).
    • Plate 2: Incubate at 44°C for 16-24 hours (for E. coli).
  4. Colony Observation:
    • Identify colony color and medium color for results.

Interpretation of Results
