The Liofilchem TERGITOL 1.5% Supplement is a sterile liquid additive used in conjunction with Tergitol Agar Base (ref. 610184 / 620184). When incorporated into the agar base, this supplement creates a highly selective medium for the detection, enumeration, and identification of Escherichia coli and coliform bacteria in water samples.     • Mix thoroughly to ensure even distribution of the supplement.
  2. Additional Supplementation:
    • Add 2.5 mL of TTC 1% Supplement (ref. 80300 / 80430) to 1 litre of base medium after sterilization.
  3. Dispensing:
    • Pour the prepared medium into sterile Petri dishes or final containers.
  4. Inoculation:
    • Use membrane filtration or direct plating techniques to inoculate the medium.
  5. Incubation:
    • For total coliforms: Incubate at 37°C for 24 hours.
    • For E. coli: Incubate at 44°C for 16-24 hours.

Interpreting Results

  • Typical Colony Morphology:
    • Escherichia coli: Yellow-orange colonies on a yellow medium.
    • Enterobacter/Klebsiella: Yellow colonies on a yellow medium.
    • Proteus/Salmonella/Shigella: Dark red colonies on a green medium.
  • Use further confirmatory tests, if necessary, for precise identification.
