Liofilchem® Amies Transport Medium w/o Charcoal is a semi-solid, phosphate-buffered medium designed to collect, transport, and preserve microbiological specimens. It maintains the viability of microorganisms during transport without promoting significant growth. This medium is suitable for various clinical and environmental microbiology applications and is particularly effective for maintaining anaerobic bacteria viability.
Key Features
- Non-Nutritive, Phosphate Buffered Medium
- Preserves specimen integrity during transport.
- Reduced Environment
- Contains sodium thioglycollate to lower redox potential, enhancing anaerobic bacteria survival.
- Semi-Solid Formulation
- Facilitates specimen stability and minimizes contamination risk.
- Wide Applicability
- Suitable for various clinical specimens and microorganisms, including Haemophilus influenzae, Neisseria gonorrhoeae, Streptococcus pneumoniae, and Streptococcus pyogenes.
Typical Formula (g/l)
Component | Concentration |
---|---|
Sodium Chloride | 3.0 |
Calcium Chloride | 0.1 |
Potassium Chloride | 0.2 |
Magnesium Chloride | 0.1 |
Monopotassium Phosphate | 0.2 |
Disodium Phosphate | 1.15 |
Sodium Thioglycollate | 1.0 |
Agar | 4.0 |
Final pH: 7.2 ± 0.2 at 25°C
Preparation
- Suspend 9.8 g of powder in 1 liter of distilled or deionized water.
- Heat to boiling with frequent stirring until completely dissolved.
- Dispense into final containers.
- Sterilize in an autoclave at 121°C for 15 minutes.
Instructions for Use
- Sample Collection
- Use sterile swabs to collect specimens. Insert one-third of the swab into the medium. Cut the stick, screw down the cap to immerse the swab fully.
- Ensure the cap is tightly secured, and keep the medium cool during transport.
- Storage During Transport
- Store the sample under cool conditions and inoculate onto appropriate culture media within 24 hours for optimal results.
Quality Control Data
Control Strain | Inoculum (CFU) | Incubation | Reaction |
---|---|---|---|
Haemophilus influenzae ATCC® 49766 | 50-100 | Microaerobic, 18-24 h, 35 ± 2°C | Good growth |
Neisseria gonorrhoeae ATCC® 19424 | 50-100 | Microaerobic, 18-24 h, 35 ± 2°C | Good growth |
Streptococcus pneumoniae ATCC® 49619 | 50-100 | Microaerobic, 18-24 h, 35 ± 2°C | Good growth |
Streptococcus pyogenes ATCC® 19615 | 50-100 | Microaerobic, 18-24 h, 35 ± 2°C | Good growth |
Storage and Stability
- Dehydrated Powder: Store at 10-30°C in a dry environment, tightly sealed in its original container.
- Prepared Tubes: Store at 2-8°C, protected from light.
- Shelf Life: 4 years for powder; 1 year for prepared medium.
Limitations
- Contaminants may grow during extended transport.
- Specimens should be inoculated into appropriate media promptly upon arrival.
- Not suitable for all fastidious organisms.
- Neisseria gonorrhoeae survives well for 6-12 hours but viability decreases significantly after 24 hours.
