Flow Cytometry Protocol & Application

1. Sample Preparation & Staining

• Cells: Human PBMCs or cultured immune cells.
• Fixation & Permeabilization: Use Intracellular Fixation & Permeabilization Buffer Set (88-8824-00).
• Recommended Isotype Control: Rat IgG1 kappa (eBRG1), PerCP-eFluor™ 710.
• Staining Procedure:
  1. Culture and stimulate PBMCs under Th2-polarizing conditions.
  2. Restimulate with Cell Stimulation Cocktail (00-4975-03) before intracellular staining.
  3. Stain with Anti-Human CD4 FITC and Anti-Human IL-10 PerCP-eFluor™ 710.
  4. Fix and permeabilize for intracellular cytokine detection.
