High-Sensitivity Sandwich ELISA for Quantitative Detection of Ceramide in Human Samples

The Human Ceramide ELISA Kit is a high-precision, sandwich-based ELISA system designed for the quantitative measurement of ceramide concentrations in human serum, plasma, cell culture supernatant, and other biological fluids. This kit utilizes a pre-coated microplate with a specific antibody to human ceramide and provides a reliable and reproducible method for monitoring bioactive lipid signaling molecules involved in apoptosis, metabolic diseases, neurodegeneration, and inflammation.

The assay is based on HRP-conjugated detection antibodies and TMB substrate, offering a sensitivity of 1 pg/mL and a dynamic detection range of 10–200 pg/mL. All reagents are stored at 2–8°C, and the kit is validated for use over a 6-month period.

Key Features & Benefits

Ultra-High Sensitivity – Detection limit as low as 1 pg/mL

Wide Assay Range – 10 to 200 pg/mL for accurate quantification of ceramide

Validated for Multiple Sample Types – Serum, plasma, culture media, tissue lysates, urine, CSF

Pre-Coated Plate Format – Minimizes variation and handling errors

Comprehensive Kit Contents – Includes ready-to-use reagents and standard curve

Reproducible Performance – Intra-assay CV <10%; Inter-assay CV <12%

Research Use Only – Designed for academic and pre-clinical studies

Applications

  • Lipid Metabolism Studies – Investigating ceramide’s role in sphingolipid signaling
  • Neurodegeneration & Apoptosis Research – Monitoring ceramide levels in CNS-related studies
  • Diabetes & Cardiovascular Research – Ceramide profiling in metabolic disease models
  • Cell Signaling Pathway Analysis – ELISA-based lipid biomarker quantification
  • Pre-clinical Screening and Assay Development – Use in drug response and disease model validation

Assay Workflow Overview

  1. Dilute standards to prepare a six-point standard curve (270–15 pg/mL)
  2. Add 50 µL of standard or sample (diluted 1:5) to each well
  3. Incubate at 37°C for 30 minutes, then wash plate 5 times
  4. Add HRP-conjugate, incubate again at 37°C
  5. Add Chromogen A & B, incubate 15 minutes in the dark
  6. Add stop solution, read absorbance at 450 nm within 15 minutes
  7. Quantify results using standard curve and dilution factor

The Human Ceramide ELISA Kit offers a robust, reproducible, and high-sensitivity assay for quantifying ceramide across a wide range of biological matrices. This kit is a vital tool for researchers studying lipid metabolism, cellular stress, and inflammation, and is compatible with standard microplate readers in any life science or pharmaceutical research setting.
