Fluorescence-based β-galactosidase detection in single cells by flow cytometry
The Invitrogen™ FluoReporter™ lacZ Flow Cytometry Kit provides a complete set of reagents and protocols for quantitative analysis of β-galactosidase (lacZ) activity at the single-cell level using flow cytometry. This system utilizes fluorescein di-β-D-galactopyranoside (FDG), a non-fluorescent substrate that is cleaved by β-galactosidase to yield bright green-fluorescent fluorescein.
The kit includes a license to practice patented FDG loading methods involving hypotonic shock to improve substrate entry into cells and techniques to enhance fluorescein retention in lacZ-positive cells, ensuring sensitive and specific detection.
Key Features
- Single-Cell Resolution – Detect lacZ expression in individual cells by flow cytometry
- High Sensitivity – Hypotonic shock loading increases FDG uptake and detection signal
- Improved Signal Retention – Proprietary methods reduce loss of fluorescent product from lacZ-positive cells
- Complete Kit – Includes FDG substrate, β-galactosidase inhibitor PETG, chloroquine diphosphate, and propidium iodide
- Multiplex Capability – Propidium iodide enables viability discrimination during analysis
Applications
- lacZ reporter gene detection in mammalian cells
- Quantification of transfection or transduction efficiency
- Flow cytometry–based gene expression analysis
- Cell sorting based on lacZ activity and viability
- Functional assays for gene regulation studies
