Thermo Scientific™ SfiI is a rare-cutting restriction enzyme that recognizes the sequence 5′ GGCCNNNN^NGGCC 3′ / 3′ CCGGNNNNNC^CGG 5′, producing defined DNA fragments. SfiI is unusual among restriction enzymes because it requires at least two recognition sites for efficient cleavage, which may reside on the same DNA molecule or on separate molecules.

SfiI is optimized to function at 50 °C in Buffer G, providing high fidelity and reproducibility in complex genomic workflows. As part of Thermo Scientific’s collection of conventional restriction endonucleases, it is validated within the Five Buffer System and supplied with the universal Tango buffer for double digestions. All buffers are premixed with BSA, which enhances stability and neutralizes potential contaminants.

    Key Features

    • Rare-cutter enzyme – recognizes long 13-bp sequences (GGCCNNNN^NGGCC).
    • Requires two recognition sites – for efficient cleavage; sites may be on the same or different DNA molecules.
    • High-temperature activity – optimal performance at 50 °C.
    • Five Buffer System – ensures flexibility and compatibility.
    • Universal Tango buffer – included for convenient double digestions.
    • Stabilized formulation – premixed BSA enhances enzyme activity and consistency.

    Applications

    • Molecular cloning of large or rare fragments.
    • Restriction site mapping.
    • Genomic library construction.
    • Genotyping and SNP analysis.
    • Southern blotting.
    • Restriction fragment length polymorphism (RFLP).
