The Thermo Scientific™ Exonuclease III (ExoIII) is a versatile enzyme with four catalytic activities, making it an essential tool in molecular biology. Its primary 3′→5′ exodeoxyribonuclease activity is specific for double-stranded DNA (dsDNA), enabling controlled degradation from blunt ends, 5′-overhangs, or nicks. This activity releases 5′-mononucleotides and generates long stretches of single-stranded DNA, useful for sequencing and mutagenesis applications.
In addition, ExoIII exhibits 3′-phosphatase activity, RNase H activity, and apurinic/apyrimidinic (AP) endonuclease activity, extending its applications to DNA repair studies, probe preparation, and hybrid digestion. Importantly, ExoIII is inactive on 3′-overhang ends longer than four bases, DNA with 3′-terminal C-residues, and phosphorothioate-linked nucleotides, ensuring reaction specificity.
