The Invitrogen™ Superoxide Dismutase (SOD) Colorimetric Activity Kit provides a complete, ready-to-use solution for quantifying total superoxide dismutase activity in serum, plasma, tissues, cells, and erythrocyte lysates. This species-independent assay measures the activity of all SOD isoforms, including Cu/Zn-SOD (SOD1), Mn-SOD (SOD2), and Fe-SOD.
Using a 96-well microplate format and a 450 nm colorimetric readout, the assay detects SOD activity as low as 0.044 U/mL, with a standard curve range of 1.4–92 U/mL. The kit is designed for ease of use, enabling high-throughput and reproducible results for oxidative stress and antioxidant research.

Assay Principle
The kit measures SOD activity by detecting the inhibition of superoxide-mediated substrate oxidation.
- Xanthine oxidase generates superoxide in the presence of oxygen.
- Superoxide reacts with a colorless substrate in the detection reagent, producing a yellow product measured at 450 nm.
- SOD in the sample scavenges superoxide, reducing product formation. Higher SOD activity results in lower absorbance values.
