Sensitive fluorescence-based detection of ROS in live cells and fresh tissue samples
The Invitrogen™ Reactive Oxygen Species (ROS) Fluorometric Assay Kit is a highly sensitive, quantitative assay for detecting and measuring ROS in fresh tissue samples and live cultured cells. ROS, such as superoxide anion, hydroxyl radical, and hydrogen peroxide, are critical mediators in both physiological signaling pathways and pathological processes including oxidative stress, inflammation, aging, and apoptosis.
The assay employs 2′,7′-dichlorodihydrofluorescein diacetate (DCFH-DA), a non-fluorescent, cell-permeable probe. Once inside the cell, DCFH-DA is hydrolyzed by intracellular esterases to form dichlorodihydrofluorescein (DCFH), which is then oxidized by ROS into highly fluorescent dichlorofluorescein (DCF). The resulting fluorescence signal (Ex/Em = 502/525 nm) is directly proportional to the ROS concentration in the sample.
