Thermo Scientific™ BL21(DE3) Competent Cells are chemically competent E. coli cells that are optimized for the expression of heterologous genes, especially those requiring the T7 RNA polymerase system. These cells are engineered with the lambda DE3 prophage, which carries the T7 RNA polymerase gene under control of the lacUV5 promoter. This setup allows for the inducible expression of T7 RNA polymerase with IPTG, enabling high-level expression of recombinant proteins in E. coli.
BL21(DE3) cells are genetically modified to reduce protein degradation by lacking the lon protease and the outer membrane protease OmpT, which helps preserve the integrity of expressed recombinant proteins.
