The Thermo Scientific™ Oxoid™ Urea Broth Base (Dehydrated) is a liquid modification of Christensen medium, specifically designed to differentiate urease-producing Enterobacteriaceae from Salmonella and Shigella species during routine microbiological testing of rectal swabs and fecal samples. This medium offers an efficient and rapid method for identifying urease-positive organisms such as Proteus spp., saving time and resources in diagnostic workflows.
Key Features
- Efficient Differentiation:
- Quickly differentiates urease-positive Proteus spp. from Salmonella and Shigella groups, reducing unnecessary follow-up testing.
- Liquid Medium for Easy Handling:
- The fluid formulation allows inoculation with a Pasteur pipette, simplifying workflows and reducing contamination risks.
- Rapid Results:
- Positive urease reactions are visible within 2-5 hours at 35°C, enhancing laboratory efficiency.
- Reliable and Clear Detection:
- Provides distinct color changes for easy interpretation of urease activity.

Applications
- Clinical Microbiology:
- Differentiates urease-producing Enterobacteriaceae, such as Proteus, from non-urease producers like Salmonella and Shigella in fecal and rectal samples.
- Food Microbiology:
- Detects urease activity in isolates from foodborne samples for quality control.
- Environmental Testing:
- Identifies urease-positive organisms in water and soil samples.
- Research:
- Studies bacterial urease production and metabolic pathways.
Preparation Instructions
- Suspend 9g of Urea Broth Base powder in 1 liter of distilled water.
- Heat gently to dissolve completely.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Cool to 50°C, and aseptically add 40% Urea Solution (SR0020K) to a final concentration as directed.
- Mix thoroughly and dispense into sterile containers.
Advantages
- Time-Saving: Detects urease-positive organisms rapidly, within 2-5 hours.
- Cost-Effective: Reduces unnecessary testing by eliminating non-target organisms.
- Easy to Use: Liquid medium is simple to inoculate and handle with minimal contamination risk.
- Reliable Results: Clear-cut color changes ensure accurate interpretation of urease activity.
