Gold-standard chromogenic substrate for β-galactosidase detection

Invitrogen™ X-Gal is a widely used chromogenic substrate for β-galactosidase detection in molecular biology, histochemistry, and cell biology applications. Upon enzymatic hydrolysis by β-galactosidase, X-Gal is cleaved to produce galactose and 4-chloro-3-bromo-indigo, an insoluble dark blue precipitate that marks the site of enzymatic activity.

This substrate is cell-permeant and compatible with fixed cell and tissue samples, allowing detection even after fixation with glutaraldehyde. X-Gal is the gold-standard reagent for lacZ reporter assays, blue/white colony screening, and β-galactosidase-based immunohistochemical techniques.

    Key Features

    • High-contrast chromogenic detection – Produces an intense blue precipitate at enzymatic activity sites
    • Cell-permeant – Suitable for live or fixed cell applications
    • Fixation-compatible – Retains β-galactosidase activity after glutaraldehyde fixation
    • Versatile – For histochemistry, molecular biology, and immunohistochemical assays
    • Highly Specific – β-galactosidase selectively hydrolyzes β-D-galactopyranosides with minimal cross-reactivity

    Applications

    • lacZ reporter gene assays in eukaryotic and prokaryotic systems
    • Blue/white colony screening in molecular cloning workflows
    • Histochemical β-galactosidase staining in cells and tissues
    • Immunohistochemistry with β-galactosidase-conjugated detection reagents
    • Characterization of glycosidase activity in microorganisms and mammalian organelles
