Invitrogen™ X-Gal is a widely used chromogenic substrate for β-galactosidase detection in molecular biology, histochemistry, and cell biology applications. Upon enzymatic hydrolysis by β-galactosidase, X-Gal is cleaved to produce galactose and 4-chloro-3-bromo-indigo, an insoluble dark blue precipitate that marks the site of enzymatic activity.
This substrate is cell-permeant and compatible with fixed cell and tissue samples, allowing detection even after fixation with glutaraldehyde. X-Gal is the gold-standard reagent for lacZ reporter assays, blue/white colony screening, and β-galactosidase-based immunohistochemical techniques.
