Qualitative ELISA for Simultaneous Detection of HIV-1/2 Antibodies and p24 Antigen in Human Samples

The Human HIV (1+2 Antibodies Plus p24 Antigen) ELISA Kit is a colorimetric, sandwich-based enzyme-linked immunosorbent assay (ELISA) designed for the qualitative detection of antibodies against HIV-1 and HIV-2 (IgG, IgM, IgA) and HIV p24 core antigen in human serum, plasma, tissue homogenates, and other biological fluids.

This combined antibody/antigen detection format improves diagnostic window coverage by enabling detection of early HIV infection (via p24 antigen) as well as established immune response (via HIV antibodies). The assay is highly sensitive and specific, uses a 450 nm optical density readout, and provides a reliable platform for research studies involving HIV exposure, vaccine development, and immunopathology.

Research Use Only (RUO) – Not for use in diagnostic or therapeutic applications.

    Key Features & Benefits

    • Dual Detection Capability – Simultaneous detection of HIV-1/2 antibodies and p24 antigen
    • Colorimetric, Sandwich ELISA – Enables qualitative measurement via absorbance at 450 nm
    • Comprehensive Sample Compatibility – Suitable for serum, plasma, and tissue homogenates
    • High Sensitivity & Specificity – Ideal for detecting both early and chronic stages of HIV infection
    • Stable & Reliable – Valid for 6 months with <5% activity loss under proper storage
    • Ready-to-Use Kit Format – Includes pre-coated plates, standards, detection reagents, substrates, and buffers

    🧪 Assay Principle

This is a sandwich ELISA. A monoclonal antibody specific to HIV p24 antigen or antibodies is pre-coated on a 96-well microplate. Samples and standards are added, followed by incubation with biotinylated Detection Reagents A and B. After washing, TMB substrate is added, resulting in a colorimetric change. The intensity of the yellow signal (after adding stop solution) is measured at 450 nm, and results are interpreted by comparing OD values to a predetermined cutoff value.

    📈 Assay Procedure Overview

    1. Add Standards, Controls, and Samples to designated wells
    2. Incubate with Detection Reagent A, then Detection Reagent B
    3. Wash thoroughly after each step
    4. Add TMB substrate, incubate, then add stop solution
    5. Read OD at 450 nm within 15 minutes
    6. Interpret results based on cutoff calculation from negative control OD + 0.15
