The Invitrogen™ Amplex™ Red Glucose/Glucose Oxidase Assay Kit offers a highly sensitive and straightforward method for detecting D-glucose or glucose oxidase activity using a fluorescence microplate reader, fluorometer, or spectrophotometer. Utilizing the Amplex™ Red reagent (10-acetyl-3,7-dihydroxyphenoxazine), this kit couples the oxidation of D-glucose by glucose oxidase with the peroxidase-mediated conversion of Amplex™ Red into resorufin, a bright, stable, red-fluorescent product.
The assay enables detection of D-glucose concentrations as low as 3 μM and glucose oxidase activity as low as 0.05 mU/mL. It is suitable for continuous or endpoint measurements and is designed to minimize background autofluorescence in most biological samples.

Assay Principle
- Glucose oxidase reacts with D-glucose to produce D-gluconolactone and H₂O₂
- Horseradish peroxidase (HRP) uses H₂O₂ to oxidize Amplex™ Red reagent
- Resorufin is formed, emitting bright red fluorescence (~585 nm) proportional to glucose concentration or enzyme activity
