The Invitrogen™ Amplex™ Red Galactose/Galactose Oxidase Assay Kit provides a highly sensitive and straightforward fluorescence-based method for detecting galactose oxidase activity or galactose concentration in biological samples.

With the ability to detect as little as 2 mU/mL of galactose oxidase activity or 4 μM galactose, this assay is ideal for enzyme activity measurement, carbohydrate metabolism studies, and biochemical screening. It is compatible with fluorescence microplate readers, spectrophotometers, and fluorometers.

Assay Principle

The assay is based on the enzymatic oxidation of galactose:

1. Galactose oxidase catalyzes the oxidation of galactose at the C6 position, producing hydrogen peroxide (H₂O₂).
2. In the presence of horseradish peroxidase (HRP), H₂O₂ reacts 1:1 with the Amplex™ Red reagent to form resorufin, a bright red-fluorescent product.
3. Resorufin’s excitation/emission maxima (571/585 nm) ensure minimal background interference from autofluorescence.
