Liofilchem® Itraconazole ITC Discs (50 µg) are premium-quality antifungal susceptibility testing (AFST) discs designed for use in the Kirby-Bauer disk diffusion method on agar media suitable for yeast and fungal growth. Each disc is impregnated with 50 micrograms of Itraconazole, a broad-spectrum triazole antifungal agent used to evaluate the susceptibility of Candida spp., Cryptococcus neoformans, and Aspergillus spp. These discs are manufactured under strict ISO 9001 and ISO 13485 certified quality systems to ensure accuracy, reproducibility, and sterility for clinical and research applications in mycology.

    Key Features

    • Standardized dosage: Each disc contains 50 µg of Itraconazole for accurate and reproducible antifungal susceptibility testing.
    • Broad-spectrum azole antifungal: Effective against a range of pathogenic fungi including yeasts and filamentous fungi.
    • CLSI/EUCAST compliant: Suitable for laboratories following standardized protocols for antifungal disk diffusion testing.
    • Sterile and ready-to-use: Packaged in sealed cartridges or canisters with desiccant to maintain potency.
    • Color-coded labeling: For easy identification of disc type and concentration during testing.

    Applications and Use Cases

    1. Clinical Mycology

Used in diagnostic laboratories to determine the susceptibility of:

    • Candida albicans, C. tropicalis, C. krusei, C. glabrata
    • Cryptococcus neoformans
    • Aspergillus fumigatus, Aspergillus niger

Supports antifungal therapy decisions in cases of:

    • Oropharyngeal and esophageal candidiasis
    • Invasive aspergillosis
    • Systemic mycoses in immunocompromised patients
    • Dermatophytosis and onychomycosis (especially for Trichophyton spp.)

    2. Antifungal Resistance Surveillance

    • Monitor azole resistance trends in Candida and Aspergillus spp.
    • Detect reduced susceptibility or resistance in azole-resistant strains of Aspergillus fumigatus

    3. Research and Pharmaceutical Development

    • Comparative activity studies of Itraconazole vs other azoles (e.g., Fluconazole, Voriconazole)
    • Validation of new formulations or combination therapies
    • Mycology drug development and fungal genomics research

    Use Instructions

    1. Prepare Mueller-Hinton agar supplemented with 2% glucose and 0.5 µg/mL methylene blue (CLSI M44 standard for yeasts).
    2. Standardize fungal inoculum to a 0.5 McFarland turbidity using a densitometer or turbidity standard.
    3. Inoculate agar plates uniformly with the fungal suspension.
    4. Apply one Itraconazole disc (50 µg) using sterile forceps or a disc dispenser.
    5. Incubate at 35 ± 2 °C for 24–48 hours depending on the organism.
    6. Measure the zone of inhibition in millimeters.
    7. Interpret the results using CLSI M44-A or EUCAST interpretive guidelines for Itraconazole, if available for the tested organism.

    Storage and Shelf Life

    • Storage Conditions: 2–8 °C (refrigerated), sealed with desiccant to prevent moisture degradation.
    • Shelf Life: 24–36 months from the date of manufacture under appropriate storage.

Liofilchem® Itraconazole ITC Discs (50 µg) offer a reliable, standardized solution for antifungal susceptibility testing of clinically relevant yeasts and molds. With broad-spectrum azole activity, these discs are essential in clinical mycology laboratories, surveillance programs, and research environments. Manufactured under internationally recognized quality systems, they help support informed antifungal therapy, resistance monitoring, and drug development efforts.
