The Liofilchem® Chloramphenicol Supplement (0.05 g per vial, 10 vials per pack) is a selective lyophilized additive designed to inhibit bacterial growth in culture media, thereby facilitating the isolation and cultivation of fungi, including yeasts and molds. This supplement is commonly incorporated into media such as Rose Bengal Agar Base to enhance fungal isolation from clinical and non-clinical specimens.
Composition:
- Chloramphenicol: 0.05 g per vial
Procedure for Use:
- Reconstitution:
- Aseptically reconstitute the contents of one vial with 7 mL of a solution comprising 50% ethyl alcohol and 50% sterile distilled water.
- Mix thoroughly until the chloramphenicol is completely dissolved.
- Incorporation into Culture Medium:
- Add the reconstituted chloramphenicol solution to 500 mL of Rose Bengal Agar Base.
- Sterilize the mixture by autoclaving at 121°C for 15 minutes.
- Preparation of Agar Plates:
- Allow the sterilized medium to cool to 45–50°C.
- Mix gently to ensure uniform distribution of the supplement.
- Pour the medium aseptically into Petri dishes and allow it to solidify.
Quality Control:
To ensure the efficacy of the supplemented medium, perform quality control using standard reference strains:
Organism | ATCC® Number | Expected Growth | Colony Appearance |
---|---|---|---|
Candida albicans | ATCC® 10231 | Good | Pink colonies |
Aspergillus brasiliensis | ATCC® 16404 | Good | White mycelium with black spores |
Escherichia coli | ATCC® 25922 | Inhibited | — |
Incubate the inoculated plates at 25–30°C for 2–7 days and observe for the expected results.
Storage and Stability:
- Unopened Vials: Store at 2–8°C, protected from light, until the expiration date indicated on the label.
- Reconstituted Solution: Use immediately; do not store for future use.
Precautions:
- Handle the supplement under aseptic conditions to prevent contamination.
- Ensure complete dissolution of the supplement before adding it to the culture medium.
- Verify the sterility and performance of the prepared medium through quality control testing before use.
By following these procedures, the Liofilchem® Chloramphenicol Supplement can be effectively utilized to prepare selective media for the isolation and cultivation of fungi, aiding in accurate microbiological analyses.
