Thermo Scientific™ PMSF (Phenylmethylsulfonyl Fluoride) is a widely used serine protease inhibitor designed to protect proteins from enzymatic degradation during cell lysis, protein extraction, and purification workflows. It is particularly effective against key serine proteases such as trypsin, chymotrypsin, thrombin, and papain, making it an essential reagent in protein research laboratories.
Supplied as a high-purity white powder (>99%), PMSF offers reliable performance in biochemical and proteomics applications where targeted inhibition of protease activity is required.
Key Features
Targeted Protease Inhibition
- Inhibits serine proteases including trypsin, chymotrypsin, thrombin, and papain
- Also inhibits certain cysteine proteases (reversible with DTT)
- Suitable for selective protease inhibition strategies
Effective at Low Concentrations
- Recommended working concentration: 0.1–1 mM
- Efficient inhibition in most protein solutions
- Minimizes interference with downstream applications
High Purity and Consistency
- Purity greater than 99%
- Low moisture content (<0.1%)
- Consistent performance across batches
Flexible Preparation
- Easily prepared as stock solutions in organic solvents
- Compatible with ethanol, methanol, or isopropanol
- Suitable for rapid integration into lysis buffers
Applications
- Protein extraction and purification
- Cell lysis workflows
- Enzyme inhibition studies
- Proteomics and protein stability analysis
- Biochemical research and assay development
Mechanism of Action
PMSF irreversibly inhibits serine proteases by reacting with the active site serine residue, thereby blocking enzymatic activity. This prevents proteolytic degradation of proteins during sample preparation.
Additionally, PMSF can inhibit certain cysteine proteases, although this effect is reversible under reducing conditions (e.g., DTT treatment).
