Gibco™ Medium 199 was originally formulated for nutritional studies of chick embryo fibroblasts, and has since become a widely adopted basal medium for the cultivation of primary and non-transformed mammalian cells. Its broad applicability and well-balanced composition make it suitable for virology, vaccine production, and explant culture of tissues such as mouse pancreatic epithelium and rat lens.
This powdered version is supplied without sodium bicarbonate and HEPES, and includes L-glutamine, phenol red, and sodium pyruvate. It is designed for reconstitution and requires pH adjustment and sterile filtration before use. Medium 199 uses a sodium bicarbonate buffering system, so it should be prepared and used in a 5–10% CO₂ incubator environment.
