Assay Principle
The 660nm assay is based on the binding of a proprietary dye-metal complex to proteins under acidic conditions, which results in a color shift from reddish-brown to green. This shift is measurable at 660 nm and is proportional to protein concentration. The dye primarily interacts with basic amino acids (lysine, histidine, arginine) and some aromatic residues (tryptophan, phenylalanine).
