EZ-Link™ NHS-Biotin is a highly efficient, amine-reactive biotinylation reagent designed for covalent labeling of primary amines on proteins, peptides, antibodies, and other biomolecules. Featuring the classic NHS ester chemistry, it reacts rapidly and irreversibly with the ε-amino groups of lysine residues and the N-terminal amine of polypeptides to form stable amide bonds.
This reagent is membrane-permeable, enabling intracellular labeling, and is ideal for creating biotin-conjugates used in affinity purification, detection, and immobilization assays involving streptavidin, avidin, or NeutrAvidin™.
Key Features & Benefits
-
Efficient Amine Reactivity
Labels lysine residues and N-terminal amines on proteins, peptides, and small molecules. -
Membrane-Permeable
Suitable for intracellular biotinylation in live-cell or fixed-cell protocols. -
Short Spacer Arm
Spacer length of ~13.5 Å (native biotin valeric acid) minimizes steric interference and ensures compact conjugates. -
Permanent Labeling
Forms non-cleavable, irreversible amide bonds, suitable for long-term protein tracking and interaction studies. -
High Purity & Consistency
Manufactured under rigorous quality standards to ensure reproducibility and optimal conjugation efficiency. -
Organic Solvent Solubility
Must be initially dissolved in DMSO or DMF prior to dilution into aqueous buffers for reaction.
Applications
-
Antibody & Protein Biotinylation
Label monoclonal or polyclonal antibodies for detection in Western blotting, ELISA, flow cytometry, and IHC. -
Intracellular Labeling
Introduce biotin inside cells for tracking, proximity labeling, or microscopy-based detection. -
Bioconjugate Synthesis
Prepare biotinylated peptides or enzymes for downstream affinity capture or protein-protein interaction assays. -
Bead or Surface Conjugation
Label ligands or receptors for immobilization on streptavidin-coated surfaces or magnetic beads. -
Biotinylation for Mass Spectrometry
Enable efficient enrichment of biotinylated proteins or peptides prior to LC-MS/MS analysis.
