Invitrogen™ PFB-FDGlu is a cell-permeable, fluorescein-based substrate designed for live-cell imaging and flow cytometric analysis of lysosomal glucocerebrosidase (GCase) activity. GCase is a critical lysosomal hydrolase in glycolipid metabolism, and its deficiency is implicated in diseases such as Gaucher’s disease and Parkinson’s disease.
PFB-FDGlu contains a pentafluorobenzoylamino-modified fluorescein moiety linked to two β-D-glucopyranoside groups. Once loaded into cells, the substrate localizes to endosomal and lysosomal compartments. Upon cleavage by active GCase, it releases a bright green fluorescent dye (PFB-F; excitation/emission ~492/516 nm) that remains cell-associated, enabling specific, high-sensitivity detection of GCase activity.
