Thermo Scientific™ Pierce™ Protein Coated Plates are pre-blocked polystyrene microplates coated with immunoglobulin-binding proteins, including Protein A, Protein G, Protein A/G, and Protein L. These plates provide an effective alternative to passive antibody adsorption by helping retain antibody activity and orient antibodies for improved antigen-binding performance in plate-based assays.
The coated surfaces enable antibodies or pre-formed antibody-antigen complexes to be captured onto wells for subsequent detection using plate readers. These plates are useful for many ELISA and immunoassay workflows, although they are generally not recommended for sandwich-type ELISA formats because additional antibodies may bind to remaining protein-binding sites and cause false positives.
Key Features
| Feature | Details |
|---|---|
| Immunoglobulin-Binding Surface | Available with Protein A, Protein G, Protein A/G, or Protein L coating |
| Improved Antibody Orientation | Binds antibodies in a way that helps preserve antigen-binding capacity |
| Reduced Loss of Activity | Helps avoid activity loss that may occur with passive adsorption |
| Pre-Blocked Plates | Supplied pre-blocked with SuperBlock™ Blocking Buffer |
| Consistent Coating | Designed for minimal well-to-well variation, typically less than 5% |
| Antibody Binding Capacity | Binds approximately 2–5 pmol antibody per well, based on rabbit IgG |
| Multiple Detection Formats | Available in clear, black, and white plate formats |
Applications
Pierce™ Protein Coated Plates are suitable for:
- ELISA and immunoassay workflows
- Antibody capture assays
- Immobilization of antibodies without prior purification
- Antibody-antigen complex capture
- Plate-based protein interaction studies
- Fc-mediated antibody orientation assays
- Fab, ScFv, and kappa light-chain binding workflows using Protein L plates
- Colorimetric, fluorescence, and chemiluminescence detection formats
- Single-antibody assay workflows
Protein Coating Selection Guide
| Coating | Binding Characteristics | Typical Use |
|---|---|---|
| Protein A | Strongly binds IgG from human, rabbit, guinea pig, pig, dog, rhesus monkey, and mouse IgG2a, IgG2b, IgG3 | Fc-mediated antibody capture |
| Protein G | Strongly binds IgG from many species, including human, mouse, rabbit, sheep, and goat | Broad IgG capture with low cross-reactivity to other antibody classes |
| Protein A/G | Combines Protein A and Protein G binding specificities | Broad IgG species compatibility |
| Protein L | Binds kappa light chains and all immunoglobulin classes including IgG, IgM, IgA, IgE, and IgD | Fab, ScFv, and kappa light-chain binding workflows |

Usage Note
These plates are recommended for single-antibody assays. For multiple-antibody workflows such as sandwich ELISA, unoccupied binding sites may capture subsequent antibodies and produce false-positive results.
Storage & Handling
Upon receipt, store plates at 4 °C in unopened pouches. Once opened, place unused plates in a resealable bag and store desiccated at 4 °C. Plates are shipped at ambient temperature.
