Broad-Spectrum β-Lactam Antibiotic for Microbiology & Cell Culture Applications

Sigma-Aldrich Penicillin G Sodium Salt (BioReagent, Cell Culture Grade) is a high-purity β-lactam antibiotic widely used in microbiology, molecular biology, and mammalian cell culture. It is effective against Gram-positive bacteria by inhibiting cell wall synthesis, making it an essential reagent for bacterial selection, contamination prevention, and antimicrobial studies.

With potency ≥1477 units per mg, this water-soluble antibiotic is ideal for cell culture media supplementation, antibiotic resistance research, and bacterial inhibition assays.

Key Features & Benefits

High-Purity BioReagent Grade – Ensures consistent performance in cell culture & microbiology applications

Broad-Spectrum β-Lactam Antibiotic – Effective against Gram-positive bacteria, some Gram-negative bacteria, and anaerobes

Mammalian Cell Culture-Compatible – Used for contamination prevention in cell cultures

Mode of Action: Inhibits Cell Wall Synthesis – Blocks peptidoglycan cross-linking by binding to PBPs (Penicillin-Binding Proteins)

Highly Soluble in Water (100 mg/mL) – Ensures easy preparation of antibiotic stock solutions

Long-Term Stability – Retains activity when stored at -20°C for extended periods

Validated for Research & Drug Development – Used in bacterial selection, antibiotic resistance studies, and pharmaceutical formulations

Mode of Action & Resistance

Mechanism of Action:

  • Penicillin G binds to penicillin-binding proteins (PBPs), inhibiting peptidoglycan cross-linking in bacterial cell walls.
  • This results in cell wall weakening, osmotic instability, and bacterial lysis.
  • It is most effective against actively growing bacterial cells.

Mode of Resistance:

  • Bacterial β-lactamases hydrolyze Penicillin G, rendering it inactive.
  • Some bacteria alter PBPs, reducing antibiotic binding and resistance development.
  • Resistance is common in Staphylococcus aureus, Enterococcus faecalis, and some Gram-negative bacteria.

Antimicrobial Spectrum:

Gram-Positive Bacteria (Streptococcus spp., Staphylococcus spp., Bacillus spp.)

Gram-Negative Bacteria (Neisseria meningitidis, Haemophilus influenzae)

Anaerobic Bacteria (Clostridium spp., Actinomyces spp.)

Applications

🔬 Microbiology & Bacterial Culture – Prevents contamination in bacterial media & culture systems

🦠 Mammalian Cell Culture Studies – Used in cell culture media for contamination control

⚗️ Antibiotic Resistance Research – Evaluates Penicillin G susceptibility in bacterial strains

🧪 Pharmaceutical & Drug Development – Used in antibiotic formulation & bacterial inhibition assays

🏭 Food & Beverage Safety Testing – Screens for penicillin-resistant bacterial contamination

Preparation & Storage Guidelines

1️⃣ Preparation of Penicillin G Stock Solution

  • Dissolve 100 mg/mL in sterile water or PBS.
  • Filter sterilize before use (do not autoclave).
  • Store at 2–8°C (short-term) or -20°C (long-term) for extended stability.

2️⃣ Recommended Working Concentrations

  • Bacterial Selection & Growth Inhibition: 10–100 µg/mL
  • Mammalian Cell Culture Protection: 50–100 U/mL

3️⃣ Storage & Handling

  • Keep tightly sealed in a dry, ventilated environment.
  • Protect from moisture & high temperatures to prevent degradation.
  • Avoid repeated freeze-thaw cycles for optimal stability.
