Miltenyi LS Separation columns

Miltenyi Biotec LS Separation Columns are high-performance magnetic separation columns designed for the positive selection of cells and for the depletion of strongly magnetically labeled cells in research and clinical laboratory workflows. They are optimized for use with MACS® MicroBead technology, enabling rapid, gentle, and highly efficient cell separation while preserving cell viability and functionality.

LS Columns are particularly well suited for applications where cells are minimally labeled, allowing downstream antibody staining, functional assays, or further processing without epitope masking or cellular stress.

Principle of Operation

The matrix of LS Columns is composed of ferromagnetic spheres coated with a cell-friendly surface. When the column is placed inside a MACS® Separator, the spheres amplify the magnetic field by approximately 10,000-fold, creating a high-gradient magnetic field within the column.

This amplified field enables:

• Efficient retention of magnetically labeled cells, even when labeling density is low
• Free passage of unlabeled cells through the column
• Minimal physical stress, as cells are held in suspension rather than binding to the matrix

The large inter-sphere spacing, several times larger than most primary and cultured cells, allows cells to flow freely, prevents clogging, and enables efficient sterile washing while avoiding cell aggregation.
