Result Interpretation for Liofilchem® EnteroPluri-Test
The results of the Liofilchem® EnteroPluri-Test are interpreted by evaluating the color changes in each of the 12 sectors, which correspond to specific biochemical reactions. Each color change indicates a positive or negative result for the respective reaction. A unique biochemical profile code is then generated, which is used to identify the microorganism.
Step-by-Step Interpretation Process
- Observation of Color Changes:
- After 18–24 hours of incubation at 36 ± 1°C, observe the 12 sectors of the EnteroPluri-Test.
- Compare the colors in each sector with the provided reference chart.
- Record Positive and Negative Reactions:
- Assign a value of positive (+) for color changes indicating a specific reaction and negative (-) for no color change.
- For example:
- Yellow in the Glucose Fermentation sector indicates acid production (positive reaction).
- Red in the Indole Production sector after adding Kovac's reagent indicates a positive indole reaction.
- Biochemical Reaction Profile:
- Divide the test results into five groups of reactions based on the sectors.
- Assign a numerical value to each positive reaction within a group. The numerical values are typically weighted (e.g., 1, 2, 4) and added to generate a group code.
- Generate a Biochemical Profile Code:
- Combine the group codes in order to create a five-digit profile code representing the biochemical reactions of the microorganism.
- Identify the Microorganism:
- Use the EnteroPluri-Test Code Book to match the biochemical profile code with the corresponding microorganism.
- The code book contains an extensive list of Enterobacteriaceae and other Gram-negative bacteria with their respective codes.
Example of Interpretation
Sector | Reaction | Color Change | Result |
---|---|---|---|
Glucose Fermentation | Acid production | Yellow | Positive (+) |
Gas Production | Gas release | Gas bubbles in the medium | Positive (+) |
Lysine Decarboxylation | Decarboxylation of lysine | Purple | Positive (+) |
Ornithine Decarboxylation | Decarboxylation of ornithine | Purple | Positive (+) |
H2S Production | Hydrogen sulfide production | Black precipitate | Positive (+) |
Indole Production | Tryptophan metabolism | Red after adding Kovac’s | Positive (+) |
Biochemical Profile Code:
- Positive reactions in a group are assigned numerical values (e.g., 4, 2, 1).
- For instance, if a group has reactions 4 (positive), 2 (positive), and 1 (negative), the sum is 6 for that group.
- Combining the results from all five groups generates the final profile code.
Example Code: 65231
Microorganism Identified: Refer to the EnteroPluri-Test Code Book to match the code with the bacterial species.
Additional Notes
- Positive and Negative Controls: Perform tests with known positive and negative control organisms to validate results.
- Reagent Addition: For certain tests (e.g., indole and Voges-Proskauer), reagents must be added post-incubation to confirm results.
