The Invitrogen™ IFN gamma Monoclonal Antibody (XMG1.2), conjugated to Brilliant Violet™ 711 (BV711), is a premium reagent designed for intracellular detection of mouse IFN gamma using flow cytometry. IFN gamma, a key cytokine produced by activated T cells, NK cells, and B cells, plays a critical role in immune regulation, macrophage activation, and antiviral responses. This antibody is validated for intracellular staining, offering bright fluorescence for clear and reliable detection.

Key Features:

  1. Target and Specificity:
    • Recognizes mouse IFN gamma, a cytokine involved in immune regulation, antiviral activity, and macrophage activation.
  2. Clone and Conjugate:
    • Clone: XMG1.2, a neutralizing antibody known for high specificity and reliability.
    • Conjugate: Brilliant Violet™ 711, a tandem dye offering bright fluorescence with excitation at 407 nm and emission at 713 nm.
  3. Validated Applications:
    • Optimized for intracellular staining in flow cytometry.
  4. Bright and Stable Fluorescence:
    • BV711 provides high signal intensity, minimizing background and ensuring clear separation of positive and negative populations.
  5. Pre-Titrated and Ready-to-Use:
    • Supplied at a concentration of 0.2 mg/mL, with recommended usage of 1 µg per test.

    • Detect intracellular IFN gamma expression in mouse T cells, NK cells, and other immune cells.
    • Ideal for use in multicolor staining panels.
  2. Th1 Research:
    • Study IFN gamma's role in Th1 differentiation and immune responses.
  3. Macrophage Activation Studies:
    • Investigate IFN gamma's effects on macrophage activation and cytokine production.
  4. Cytokine Profiling:
    • Analyze IFN gamma production in immune cells in response to antigen or mitogen stimulation.
  5. Immunotherapy Research:
    • Explore IFN gamma's involvement in tumor immunity and therapeutic interventions.

Key Benefits:

  1. Bright Fluorescence:
    • BV711 offers superior brightness and stability for clear identification of IFN gamma-positive cells in flow cytometry.
  2. Minimal Background:
    • High specificity of the XMG1.2 clone ensures accurate results with minimal non-specific binding.
  3. Reproducibility:
    • Pre-titrated and validated for consistent performance across experiments.
  4. Multicolor Panel Compatibility:
    • BV711's spectral properties make it highly compatible with other fluorochromes in multicolor staining panels.
  5. User-Friendly:
    • Ready-to-use liquid formulation simplifies experimental workflows.

Usage Recommendations:

  1. Sample Preparation:
    • Stimulate mouse splenocytes with appropriate antigens or mitogens to induce IFN gamma production.
  2. Staining Protocol:
    • Use the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00) for optimal intracellular staining.
    • Add 1 µg of antibody per 100 µL of cell suspension.
  3. Flow Cytometry:
    • Analyze samples using a flow cytometer equipped with a violet laser (407 nm).
  4. Fixation:
    • Fix samples in IC Fixation Buffer to preserve fluorescence for up to 3 days.
  5. Minimize Non-Specific Binding:
    • Use Brilliant Stain Buffer™ to reduce polymer dye interactions when combining multiple Brilliant Violet™ reagents.

Storage and Handling:

  • Store at 4°C in the dark to protect the Brilliant Violet™ 711 fluorochrome.
  • Avoid freezing, as freezing may degrade the antibody or fluorochrome.

Target Information:

  • IFN gamma:
    • A dimeric cytokine (~45 kDa) secreted by activated T cells, NK cells, and B cells.
    • Plays a critical role in macrophage activation, MHC class I and II expression, and antiviral responses.
    • Acts via the JAK-STAT signaling pathway, synergizing with other cytokines to regulate immune responses.
