Assay Procedure (Overview)

1. Add 10 µL of sample + 40 µL of sample diluent to test wells
2. Add 50 µL of positive/negative controls to control wells
3. Add 100 µL HRP-Conjugate to each well
4. Incubate 60 min at 37°C
5. Wash 5× with diluted wash buffer
6. Add 50 µL each of Chromogen A & B, incubate 15 min (avoid light)
7. Add 50 µL Stop Solution
8. Read OD at 450 nm within 15 minutes
9. Interpret results using calculated cutoff:
   • Negative: OD < Cutoff
   • Positive: OD ≥ Cutoff
