The Human MDA ELISA Kit is a high-sensitivity sandwich ELISA designed for the quantitative detection of malondialdehyde (MDA) in human serum, plasma, urine, culture media, tissue homogenates, and other biological fluids. Malondialdehyde is a biomarker of oxidative stress and lipid peroxidation, commonly elevated in pathological states including cardiovascular disease, cancer, inflammation, and neurodegeneration.

This kit offers robust performance, broad sample compatibility, and precise quantification, making it ideal for academic, preclinical, and translational research settings.

Key Features

  • Assay Format: Sandwich ELISA
  • Detection Range: 7 – 400 ng/mL
  • Sensitivity: 1.5 ng/mL
  • Sample Volume Required: 10 μL
  • Detection Wavelength: 450 nm
  • Assay Duration: ~2.5 hours
  • Storage Conditions: 2–8 °C
  • Shelf Life: 6 months
  • Precision:
    • Intra-assay CV <10%
    • Inter-assay CV <12%

Assay Principle

The ELISA plate is pre-coated with a monoclonal antibody specific to MDA. Standards or test samples are added and bind to the immobilized antibody. An HRP-conjugated detection antibody is then added, forming a sandwich complex. TMB substrate is introduced and converted to a blue product, which turns yellow upon the addition of stop solution. Absorbance is read at 450 nm, and MDA concentration is calculated using a standard curve.

Assay Procedure Summary

  1. Standard Dilution: Prepare six standards (540 → 30 ng/mL).
  2. Add 40 μL sample diluent + 10 μL sample to wells (except blank).
  3. Incubate 30 minutes at 37°C, sealed.
  4. Wash plate using diluted wash buffer.
  5. Add 50 μL HRP-conjugate, incubate again.
  6. Wash and add 50 μL each of Chromogen A & B.
  7. Incubate 15 minutes in the dark at 37°C.
  8. Add 50 μL stop solution. Measure OD at 450 nm within 15 minutes.

Performance Summary

ParameterSpecification
Assay Range7 – 400 ng/mL
Sensitivity1.5 ng/mL

Inter-assay CV<12%

The OD values are plotted on a log-log scale. Multiply final values by the dilution factor (typically ×5) to obtain original sample concentrations.

The Human Malondialdehyde (MDA) ELISA Kit (SL1135Hu) is a valuable tool for researchers studying oxidative stress, lipid peroxidation, and related metabolic or inflammatory conditions. With excellent sensitivity, robust performance, and broad sample compatibility, this kit provides accurate and reproducible results for quantifying MDA in both clinical and experimental research settings.
