🧪 Assay Procedure Summary

1. Add 10 μL sample + 40 μL sample diluent to test wells
2. Add 50 μL positive/negative control to designated wells
3. Add 100 μL HRP-conjugate reagent and incubate 60 min at 37°C
4. Wash plate 5× with diluted wash buffer
5. Add 50 μL each of Chromogen A and B, incubate 15 min at 37°C (protected from light)
6. Add 50 μL Stop Solution; observe color change from blue to yellow
7. Read OD at 450 nm within 15 minutes

📈 Result Interpretation

• Positive Result: Sample OD ≥ (Average Negative Control OD + 0.15)
• Negative Result: Sample OD < (Average Negative Control OD + 0.15)
• Validity Criteria:
  • Positive control OD ≥ 1.00
