The Human 8-Isoprostane (8-ISO) ELISA Kit is a high-sensitivity enzyme-linked immunosorbent assay designed for the quantitative measurement of 8-Isoprostane levels in human biological samples. 8-Isoprostane is a stable biomarker of oxidative stress and lipid peroxidation, making it an essential indicator in studies related to inflammation, cardiovascular diseases, neurodegenerative disorders, metabolic syndromes, and environmental toxicology.
This kit utilizes a solid-phase sandwich ELISA format with microplate wells pre-coated with antibodies specific to human 8-Isoprostane. During the assay, the antigen present in samples binds to immobilized antibodies, followed by detection with horseradish peroxidase (HRP)-conjugated antibodies. Upon addition of chromogenic substrate, a colorimetric reaction develops that is directly proportional to the concentration of 8-Isoprostane present. Absorbance is measured at 450 nm using a microplate reader, allowing precise quantification through comparison with a standard calibration curve.
The kit is optimized for use with multiple sample types including serum, plasma, cell culture supernatants, tissue extracts, urine, and other biological fluids. With excellent sensitivity, reproducibility, and reliability, this assay provides a robust tool for biomarker research in clinical, academic, and pharmaceutical laboratories.
Applications
- Oxidative stress biomarker analysis
- Cardiovascular and metabolic disease research
- Inflammation and lipid peroxidation studies
- Neurodegenerative disease research
- Toxicology and environmental health studies
- Cell culture and molecular biology research
Principle of the Assay
The assay is based on the sandwich ELISA technique:
- Microplate wells are pre-coated with anti-human 8-Isoprostane antibodies.
- Samples and standards are added, allowing antigen binding.
- HRP-conjugated detection antibody binds to the antigen-antibody complex.
- Chromogenic substrate (TMB) produces a measurable color reaction.
- Stop solution terminates the reaction, producing a yellow color.
- Absorbance is measured at 450 nm and correlated to antigen concentration using a standard curve.
