Fluorescent β-galactosidase substrate with large Stokes shift for sensitive enzyme activity detection
Invitrogen™ DDAO galactoside is a highly sensitive β-galactosidase substrate that produces a fluorescent hydrolysis product detectable in the far-red spectrum. Upon cleavage by β-galactosidase, DDAO galactoside yields a hydrolysis product with excitation/emission maxima of ~645/660 nm, making it ideal for use with 633 nm laser excitation. The substrate itself is also fluorescent (excitation/emission ~460/610 nm), and the ~200 nm spectral shift between the substrate and product allows for clear spectral separation during detection.
This property enables simultaneous monitoring of substrate and product, reducing interference and enhancing sensitivity in multi-parameter fluorescence assays. DDAO galactoside is particularly suited for flow cytometry, fluorescence microscopy, and reporter gene assays where β-galactosidase is used as a marker enzyme.
