Preparation Instructions:
- Dehydrated Medium:
- Dissolve the entire content of the medium (for 5 L preparation) in distilled water.
- Heat gently while stirring to dissolve completely.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Pouring Plates:
- Allow the sterilized medium to cool to 45–50°C.
- Pour into sterile Petri dishes under aseptic conditions.
Testing Procedure:
- Sample Preparation:
- Prepare sample dilutions as required.
- Inoculation:
- Apply the sample onto the surface of the medium using either the spread plate or pour plate method.
- Incubation:
- Incubate the plates at 44°C for 18–24 hours.
