Thermo Scientific™ BcuI (also known as SpeI) is a type II restriction endonuclease that recognizes the palindromic DNA sequence A^CTAGT and cleaves between the adenine and cytosine bases. It functions optimally at 37 °C and is provided at a concentration of 10 U/μL. BcuI is not sensitive to Dam, Dcm, or CpG methylation and performs best in Thermo Scientific’s Tango Buffer, which is part of the Five Buffer System.
This enzyme is supplied with premixed BSA in the reaction buffer to enhance stability and reduce interference from contaminants, making it ideal for cloning, mapping, and RFLP workflows. Also available as a FastDigest™ BcuI version for rapid digests.
