Usage Guidelines

Preparation

  1. Suspend 35.6 g of powder in 1 liter of distilled or deionized water.
  2. Heat until completely dissolved.
  3. Dispense into final containers with Durham tubes.
  4. Autoclave at 121°C for 15 minutes.

Technique

  1. Inoculate 1 mL of sample (or serial dilutions) into a tube of Lauryl Tryptose Broth.
  2. Invert the tube to expel air from the Durham tube.
  3. Incubate at 36 ± 1°C for 24-48 hours.

Results Interpretation

  • Positive Presumptive Test: Turbidity and gas production.
  • Confirmation: Perform the indole test directly in the tube.
