How It Works

CellTrace™ CFSE (carboxyfluorescein diacetate succinimidyl ester) diffuses into live cells and binds covalently to intracellular proteins. Upon each cell division, the dye is equally distributed between daughter cells, resulting in a halved fluorescence intensity—this enables the monitoring of sequential cell divisions using flow cytometry.

Kit Components

  • 10 × vials of CellTrace™ CFSE (lyophilized powder)
  • 1 × 500 μL vial of anhydrous DMSO

Staining Protocol Highlights

  1. Reconstitute dye in DMSO.
  2. Incubate cells (in protein-free media) with 1 µL dye per 1 mL cell suspension for 20 minutes at room temperature.
  3. Quench excess dye with complete medium.
  4. Analyze via flow cytometry using 488 nm excitation and 517 nm emission settings.
