This reporter gene assay system uses 3-carboxyumbelliferyl β-D-galactopyranoside (CUG) as the substrate. Upon hydrolysis by β-galactosidase, CUG produces 7-hydroxycoumarin-3-carboxylic acid, a bright blue-fluorescent product.
